CONSTITUTION: A protoplast derived from lower hypocotyl of Japanese radish is subjected to discontinuous density gradient centrifugation using Percoll and mannitol to isolate a cytoplast. On the other hand, protoplast derived from a Brassica plant such as rapeseed is added to iodoacetoamide to carry out iodine compound treatment. Then the abovementioned cytoplast is fused with an iodine compound-treated protoplast and further the resultant fused cell is cultured to form colony and successively adventive bud. Thereafter, the adventive bud is reared and transplanted on vermiculite and rooted to provide the objective cytoplasmic hybrid plant.
