First, RIP2 is required for IL-12-mediated upregulation of IFNg, as shown in both Th1 cells and NK cells. Second, RIP2 is involved in Toll-like receptor (TLR) signaling. RIP2 knockout mice are more resistant to endotoxic shock after challenge with bacterial lipopolysaccharide (LPS), which signals through TLR4. RIP2-/- macrophages are impaired in NF-kB and SAPK activation, as well as cytokine production in response to LPS treatment. Third, RIP2 is most likely the key kinase in mediating signals through intracellular receptors such as the NOD family proteins. NOD proteins contain CARD(s) at the amino terminus, a nucleotide binding domain in the middle, and leucine rich repeats (LRR) at the carboxyl terminus. The LRR domain is involved in the recognition of intracellular microorganisms such as bacteria and parasites. The CARD of NOD proteins can directly bind to the CARD of RIP2. Intracellular microorganisms may trigger the aggregation of the NOD/RIP2 complex, and activate innate responses. RIP2-/- mice exhibit increased susceptibility to infection to the intracellular bacteria Listeria monocytogenes .
