A finishing line for forming a square back saddle stitch book. A conveyor is arranged to receive the book and to convey the book along a finishing path to a squaring station. A book clamp is disposed on opposite sides of the path and is arranged to clamp the book at a point generally adjacent to the spine. A squaring member includes a first portion transitioning toward a second portion, the second portion disposed generally in the path and is positioned to contact and flatten the spine of a book proceeding along the path.

A binding system in an imaging system includes an accumulator in which sheet material is accumulated in respective jobs to be bound. The binding system includes a binding assembly adapted and constructed to bind the sheet material together. A scoring member is connected to the binding assembly. The scoring member is adapted and constructed to form respective score patterns in sheet material received in the accumulator. The binding assembly can include a thermally low-mass heating element and a thermally high-mass cooling element. The scoring member can be connected for motion with respect to the binding assembly, for example, by forming at least one bore in the binding assembly, and at least one guide post extending from the scoring member and received for reciprocation in the at least one bore in the binding assembly. At least one resilient biasing member, for example, a coil spring surrounding the guide post, can be disposed between the scoring member and the binding assembly. The scoring member can be constructed as an elongate wedge-shaped bar, a bar having a serrated edge, or a generally linear series of pointed protrusions. The binding assembly can include a frame assembly having a first frame member mounted generally parallel to a second frame member. The heating element can be mounted on a lower surface of the frame assembly. The binding assembly can also include a cooling element having a generally parallelepipedal lower portion extending downwardly from a generally parallelepipedal upper portion. The lower portion of the cooling element can be mounted for reciprocal movement between the first and second frame members of the frame assembly. A method of binding sheet material is also set forth.

A method is provided for identifying a restriction endonuclease, which includes the steps of (a) screening a target DNA sequence for the presence of known methylase sequence motifs, (b) identifying any open reading frames which lie close to the methylase sequence motifs screened in step (a), and (c) assaying the protein products of these open reading frames for restriction endonuclease activity. Methods for identifying isoschizomers of known restriction endonucleases, which isoschizomers possess a desired physical property, such as thermostability, are also provided by the present invention, as are several novel restriction endonucleases isolated from M. jannaschii, MjaIII and MjaIV. Additionally, a gene was identified that encoded a previously observed endonuclease activity, designated MjaII. Also provided by the present invention are vectors suitable for cloning a DNA sequence encoding a cytotoxic protein, via independent transcription promotors which may be selectively controlled by several conditions. A method for producing these cytotoxic proteins using such vectors is also provided, as are stable clones of PacI and NlaIII.

Systems and methods are described for increasing the binding strength of a bound text body by increasing the spinal surface area exposed for adhesive penetration greater than an area corresponding to the product of the sheet height dimension, the sheet thickness dimension and the number of sheets. In one aspect, sheet-wise conditioning of the binding area is performed prior to the application of adhesive to increase the bond area between sheets of the text body. In another aspect, the spinal surface area exposed for adhesive penetration is increased as adhesive is applied.

A method and apparatus of in-line die-cutting in saddle signature gathering wherein signature sets are gathered on a gathering chain to form signature sets. The signature set is grasped by a pair of belts which lift the signature off the gathering line so that a portion of the signature set along the spine thereof may be removed by a die-cutting pair of plates located above the gathering line. The signature sets may be stopped as lifting and cutting occurs. After lifting and cutting the signature set is replaced on the gathering line for further processing.
