CONSTITUTION: A leaf piece after surface sterilizing treatment is laid on a sterilizing filter paper and finely cut to thin piece of 2-5mm square length. Next, said thin cut piece is dipped in fixed axenic enzyme solution and cell wall is removed. Then, said piece is put into centrifugal tube and washed 2-3 times with mannitol liquid. Thus, sedimented protoplast is taken out from the centrifugal tube and transferred into liquid medium in petri dish, set in refrigerator and treated at low temperature of 2-5°C for 24-72 hour in darkness. Further, said petri dish is received in isothermal culturing device and protoplast is cultured under low illumination (100-200 lux) by fluorescent light at 25-30°C. By said method, percentage of success in culturing of protoplast is improved and useful for utilizing of regeneration of plant body or cell fusion.
